CAR T cell therapies have reshaped immuno‑oncology, yet functional heterogeneity driven by CAR morphology and spatial organization remains challenging to assess at scale. Growing evidence shows that how CARs are distributed on the T cell surface directly influences signaling strength, cytotoxic activity, and persistence—but these features are difficult to capture with conventional flow cytometry alone.

This application note demonstrates how the BD FACSDiscover Platform, integrating spectral flow cytometry with BD CellView Image Technology, enables high‑throughput and real-time analysis of CAR distribution alongside key phenotypic and functional markers. By leveraging fluorescence imaging parameters, researchers can move beyond population‑average measurements also sort functionally relevant CAR spatial phenotypes for downstream studies.
