Gene knock-out (KO) and knock-down (KD) are the gold standard methods for validating antibody specificity. Both approaches eliminate or silence the endogenous target protein to assess antibody binding and validate specificity. KO or KD validation is usually done at the gene level using qRT-PCR/qPCR or at the protein level using Western blotting, immunocytochemistry, or flow cytometry. However, validation using only one application has potential drawbacks.
